| Abstract: | An efficient and improved micropropagation protocol was developed from different explants of 2-month-old seedlings of Simarouba glauca DC. The highest number of shoots (55.3) with 100% response was obtained from cotyledonary-node explant on MS medium fortified with 6-benzyl adenine (BA; 3.0 mg/L), kinetin (KN; 0.5 mg/L), α-naphthalene acetic acid (NAA; 0.5 mg/L) and glutamine (10 mg/L). Further, the highest rooting response (95%) with 15 roots of 5.8 cm root length was observed on MS medium supplemented with NAA (3.0 mg/L). The rooted plantlets were hardened by transferring to paper containers with soil and sand (2:1) and kept in shade for acclimatization. These plants were further transferred to the field with a survival percentage of 81.5%. |
