Authors: John, Reni; Kariyil, Bibu John; PTA, Usha; S, Surya
Abstract: Erythrina variegata L. (Fam. Fabaceae) is traditionally known for antitumour, expectorant, febrifuge, antibacterial and antioxidant activities. In the present study, we did cytotoxic evaluation of the methanol extract of E. variegata stem bark (MEV) using 3-(4,5-dimethyl thazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay in MCF-7 and MDA-MB-231 human breast carcinoma cell lines. The cytological and metabolic alterations due to MEV was evaluated using acridine orange/ethidium bromide (AO/EB) dual staining, hoechst 33258 and fluoroprobe, benzimidazol-carbocyanine iodide 5,5',6,6'-tetrachloro-1,1',3,3'-tetra ethyl (JC-1) staining. The antiapoptotic gene and protein, Bcl-2 expression was analysed using real time PCR and Western blotting, respectively. The methanol extract was investigated for the presence of various phytochemicals using gas chromatography high resolution mass spectrometry (GC-HRMS). The results revealed a dose dependent cytotoxic activity in both MDA-MB-231 and MCF-7 breast carcinoma cell lines in vitro. The plant extract was effective in inducing apoptotic cell death via mitochondrial dependent intrinsic pathway and may be considered as a potent source for isolating therapeutic molecules against cancer.

Abstract: In rodent control exercise, after successful control with rodenticides, the surviving population often rebuild up themselves rapidly due to reduced competition and high rate of reproduction. In this context, here, we investigated the potential of papaya (Carica papaya L.) seed powder as a natural reproduction inhibitor for male lesser bandicoot rat, Bandicota bengalensis (Gray and Hardwicke) in reducing the post control population rebuild up rate. Exposure of male B.bengalensis to cereal based bait containing 2, 3 and 5% papaya seed powder for 15 and 30 days durations in bi-choicelaboratory feeding tests resulted in the ingestion of total 7.14-18.15 and 13.74-41.25 g/kg body wt. of the active ingredient,respectively. Breeding performance of treated rats was 33.33 and 16.67% after treatment with 5% papaya seed powder for15 and 30 days, respectively. Autopsy of rats immediately after treatment withdrawal revealed a significant decrease inweights of testis, cauda epididymis, seminal vesicles and prostate gland. Sperm motility, sperm viability and sperm countwere found reduced and sperm abnormality increased significantly in the cauda epididymal fluid of treated rats as comparedto untreated rats. The total proteins, 17β-HSD, 3β-HSD and testosterone levels were also found decreased in treated rats ascompared to untreated rats. All the changes observed in treated rats showed partial or no recovery after 30 days of treatmentwithdrawal. The study suggests the potential of papaya seed powder in regulating fertility of male B. bengalensis which maybe exploited as a component of integrated rodent pest management.

Abstract: Soybean (Glycine max L.) is a major legume crop cultivated principally as protein rich bean. Improving quality and yield received considerable attention from researchers. Here, we explored molecular approaches to improve soybean quality and yield. In spite of recalcitrance, soybean plants successfully regenerated through complex and time consuming in vitro regeneration protocols via organogenesis and/or somatic embryogenesis and being used for transgenic development. Transformation efficiency is highly dependent on the regeneration as not all the cells transformed lead to the recovery of viable plant regeneration. Consequently, efficient in vitro regeneration found to be directly associated with the recovery of transformants. In the present study, we standardized the in vitro florigenesis using cotyledonary node with axillary bud as explant of soybean variety JS-335. Flower buds were directly induced from proximal end of the explant on Murashige-Skoog (MS) medium augmented with thidiazuron (TDZ) and α naphthalene acetic acid (NAA). TDZ proved a potential growth regulator to induce in vitro florigenesis. As a result of in vitro fertilization, pods were developed from flowers and matured within 40-45 days on hormone-free medium. Pods and seed set under in vitro conditions resemble pods and seeds produced under in vivo conditions. This pathway of in vitro florigenesis showed great potential for successful induction of in vitro flowering, which in turn can be explored in producing transgenic soybean seeds in popular Indian soybean genotype without escaping transgene.

      <title>Evaluation of sodium nitroprusside (NO donor) as pulsing solution in improving post harvest quality of gladiolus spikes</title>
      <link>http://nopr.niscpr.res.in/handle/123456789/57852</link>
      <description>Title: Evaluation of sodium nitroprusside (NO donor) as pulsing solution in improving post harvest quality of gladiolus spikes
Authors: Mittal, Isha; Jhanji, Shalini
Abstract: Gladiolus is an important commercial cut flower of emerging floriculture industry. The prime concern in&#xD;
commercialization is postharvest management of flowers as flowers are biological systems that perish after harvest. Among&#xD;
different chemicals used to improve vase life of gladiolus, nitric oxide has emerged as potent signaling molecule that can&#xD;
delay senescence and improve post harvest life of flowers. Here, we studied the potential of sodium nitroprusside (SNP) as&#xD;
pulsing solution on post harvest quality of gladiolus. Spikes of gladiolus were harvested at tight bud stage and pulsed for&#xD;
20 h with SNP solutions of different concentrations and various physiological parameters were recorded to evaluate the&#xD;
keeping quality. The pulsing solution of SNP @200 mg L-1 with sucrose and aluminium sulphate significantly improved the&#xD;
vase life whereas pulsing with SNP @400 mg L-1 adversely affected the quality parameters. Spikes pulsed with SNP&#xD;
@200 mg L-1 supplemented with sucrose and aluminium sulphate had higher membrane stability index, relative water&#xD;
content, sugar content, protein content antioxidant enzymatic activity and lower TBARS content as compared to control&#xD;
supporting improved postharvest quality. Thus, sodium nitroprusside @200 mg L-1 with sucrose and aluminium sulphate&#xD;
could be used as pulsing solution to improve the postharvest quality of spike.
