This study presents raw inulin from Dahlia sp. as substrate at shake-flask level for inulinase production from Kluyveromyces marxianus YS-1 using response surface methodology (RSM). A total of 27 fermentation runs were carried out and experimental data was analyzed using ANOVA. Multiple correlation coefficient R was 0.9970 and 0.9890 for inulinase activity and biomass yield, respectively, which justify an excellent correlation between predicted and experimental values. Model F value of 82.50 and 20.41 for inulinase activity and biomass yield, respectively, highlights significance of model. Experimental values obtained under optimal conditions were in good agreement with RSM values. A significant increase in enzyme production by 4.5 times was achieved on optimizing the media at shake-flask level using RSM as compared to enzyme production obtained by one-variableat- a-time approach using pure inulin as a substrate. Raw Dahlia inulin proved to be a better substrate for inulinase production than raw inulin from Asparagus sp.en_US
