Abstract: Four DNA extraction protocols from the fish scales of Cyprinus carpio were compared. The DNAs obtained from all the protocols were tested by PCR and the DNAs extracted using protocol 1 and 2 were also tested by restriction analysis. The results indicated that all four protocols extracted DNA suitable for PCR amplification. However, variations in the yield and purity of extracted DNA were observed between protocols. The DNA samples prepared by protocol 1 and 2 were amenable to restriction digestion by restriction enzyme HindIII and NotI. Different protocols have their own merits and shortcomings. Thus, the present study can be used as a reference for selecting suitable protocol according to the aim and need of the study undertaken.
